2 Story Setup RBK50
In process of purchasing an RBK50 for 2 story home. First story 2000 sq ft, second story 1000 sq ft. Router on first floor in center of house, satellite on second floor approximately same position but 1 floor up. Will this work or do I need to separate them horizontally as well as vertically?

Yes. 30 feet minimum is recommended in between the base router and satellie(s) to begin with depending upon building materials.
