So planning on moving to a new 3 story 2000 SQ feet house. The router can only go to one spot which is on the first floor. Question is will I have issues with signal strength? Let me know if there ...

Try it.
Reason why is you didn't give us much details.
A single router solution can cover that much area (better if its centrally located) but you might get some lower speeds at the peripheral. For some people, that lower speed isn't an issue as they don't need it. For others, they want peak performance everywhere and would add an extender or switch to a mesh system.
All we can say is try it and see how it works. It might be fine for you or you might find you need more coverage.
I would say to make sure its not in a utility room as they tend to have a lot of hvac that blocks wifi.
