Ad a 2TB drive to a RN 516 (X-RAID2) with one 4TB drive?

Not with xraid.
You'll need to change to flexraid, add the 2 tb disk and create a new volume on the 2 tb disk, ending up with 2 unprotected volumes. You must have a backup, especially if you are not using raid redundancy.
If you create 2 volumes like this you will not in future be able to change to xraid or raid1 or raid 5 without recreating a new volume and restoring data from a backup.
