Any hot-insert will wipe the drive, so if you don't want that to happen its vital that you power down the NAS before the insertion.
You can power down the NAS and try reinstalling your original 500GB drive 1 (with slot 2 empty), and then powering on. If I understand your sequence correctly that should boot. You can also try linux reader on a PC (http://www.diskinternals.com/linux-reader/). Linux reader will only read your original drive 1, it will not read drive 2.
BTW, its a good idea to label the drives by slot.
