"seems"? Actual command? Actual error message? As usual, showing
actual actions with their actual results (error messages, LED
indicators, ...) can be more helpful than vague descriptions or
interpretations. Copy+paste is your friend.
Which "the file"? Where is the file? What is the current directory?
dir
echo %cd%
If the file is not in your current directory, then you can either move
the file, change your current directory, or else specify a proper path
to the file.

I'd expect your Command Prompt shell program to start up in your
(user-specific) home directory, perhaps something like "C:\Users\fred"
(for a user named "fred"). By default, the Command Prompt program
displays this directory path in its command prompt string
("C:\Users\fred>").
Your Desktop folder would then be "C:\Users\fred\Desktop". To get
there:
cd Desktop
To get to a sub-folder named, say, "Firmware":
cd Firmware
Then a command like the following should work (or, at least, not
complain about not finding the specified file):
tftp -i 192.168.1.1 R8500-V1.0.2.130_1.0.99.chk
Alternatively, starting in "C:\Users\fred", specify a path to the
file (instead of only its leaf name) in your TFTP command. For example:
tftp -i 192.168.1.1 Desktop\Firmware\R8500-V1.0.2.130_1.0.99.chk
A Web search for terms like, say:
windows command prompt directory
should find any number of elementary guides (some better than others, no
doubt). There are many details which may or may not affect you, too,
such as using quotation marks to cope with space characters in
file/directory names. Research as needed.
