I am currently using a Comcast router that includes Wifi. Can the Nitehawk be used in combination with such a router? Is there any advantage?

yes it can. through a couple options .
1. leave the comcast router up and running. put the netgear in AP (access point) mode. This prevents a double nat. Just make sure the wireless radios are on different channels to avoid interference.
2. put the comcast router in bridge mode. It disabled its router functions (and wifi) and leaves only the netgear in control
