New DST Range Extender
Does anyone know if the new DST wireless adapter work with a Nighthawk AC3200?

Judging from the documention (manual and FAQ), the DST6501 adapter is a HomePlug AV2 Powerline adapter with a built-in Wi-Fi Access Point. It's marketed to be used with the new R7300 DST router, which must surely be a router with a built-in Powerline adapter. Since a Nighthawk AC3200 doesn't have a built-in Powerline adapter, the DST6501 won't work directly with it.
The question is whether a DST6501 will work with a regular, non-DST, HomePlug AV Powerline adapter. Hopefully, Netgear designed it to be compatible. Perhaps a moderator can check with Netgear engineering, respond here and update the FAQ.
If it turns out to be compatible, then all you need is another Powerline adapter to make it work with your Nighthawk. On the other hand, if you don't own a R7300 DST router there is limited value in getting the DST6501. The attraction of the DST adapter/router combo is the plug and play ease of setup, but that literally seems to amount to them being preconfigured with the same Powerline password. In fact, the password is not even random (it's HomePlugAV), so I'm not sure anyone would want to use it without changing it. But then you've lost the plug and play advantage and are no better off than buying regular Powerline adapters.
As for the R7300 DST, I suppose it's kinda nifty to have built-in support Powerline. The R7300 manual doesn't list the specs of its Powerline adapter. Personally, I'm fan of keeping technology components separate (within reason) for more flexibility in upgrading, but to each their own.
Note: Take this as a personal opinion based on a quick perusal of the documentation. My observations and, hence, conclusions could be mistaken.

My biggest question here is whether my existing R7000 router will be able to support DST via a firmware update or do I need a brand-new R7300 router to support DST via some sort of hardware improvement. I am guessing the answer is that I would need to be a new router although I would like an answer back from Netgear on this.
There's no way the R7000 can support DST via a firmware update. It lacks the hardware to support Powerline networking.
Since you already own the R7000, you would be better off buying a pair of Powerline adapters than upgrading to the R7300. The R7300 is just a R7000 with a built-in Powerline adapter. It looks like there are already newer Powerline adapters available that are faster than what the R7300 supports.
