CANNOT READ FILE PERMISSION DENIED

We need a lot more information on this.
At least
- network access settings for the share
- file access settings for the share
- username that cannot read the file.
There is a Reset control on the file access settings page for each share that you could try.

We are trying to access from the $ share via built-in Administrator account. Initially it connects & pulls out some GB data afterwards results in error as
cannot read file 'file name' permission denied

Are you saying that you want to back up a PC to the NAS using a built-in backup job from the PC's $ share?

Actuallly we are pulling data from Windows 10 Pro machine, which has built-in C$,D$ Drive Shares out of which C$ is getting copied but D$ is resulting an error.

As you likely know, some files/folders are locked down and can't be copied.
If it's not those, then you either need to adjust file permissions and/or sharing permissions on the PC, or change the credentials used on the ReadyNAS.
Or push the data from an account that has access, instead of pulling it.
