Fastest copy way to copy huge file from NAS to the pc?

I wouldn't use ReadyCLOUD.
Not sure what you mean be directory copy between USB. You can't connect the PC to the NAS with USB. But if you have a USB drive, you could connect it to the NAS, and back up to it with a ReadyNAS backup job.
Otherwise, you should just use gigabit ethernet. There are some copy utilities out there that will verify, you could look into those.
