You can do most of what you want with the flexraid mode. The NAS will need to format your disks - if you are using 4.2.x firmware, you can find instructions in the manual here: http://www.downloads.netgear.com/files/GDC/RNDU2000/ReadyNAS_Home_RAID4.2SW_en_29-Sep-11.pdf
Your NAS can also be switched to OS 6, and since you need to do a factory reset anyway you might want to consider doing that.

No. The NAS uses a linux file system (ext with 4.2 firmware, btrfs with 6.x firmware). Windows doesn't support either.
You can create four volumes (one per disk) with various network shares on the NAS. But the NAS software isn't designed to let you insert/remove hard drives as if they were optical disks.
