I'I just tested it and reproduced the same behavior. There is nothing "wrong with the NAS".
In my opinion, this is a bug, or an annoying limitation.
1. Inserted a USB stick in the NAS.
2. Created a backup job that points to the USB stick (USB share), with scheduled enabled.
3. Ejected the USB stick from the NAS.
4. Tried to disable the schedule. I get the error "be sure to correct errors".
5. Under Destination, "Type" is in red and value is "No longer exists".
I'm running 6.7.1-T252
Workarounds I can think of: delete the job or change the destination to a USB port instead of a USB share.

Possibly. I was mainly thinking of modifying the job temporarily to something that would allow saving, without necessarily keeping the job working. Next time the USB drive is connected, it'd be easy to quickly edit the job again.

I'd use the 127.0.0.1 trick with either the source or the destination anyway, since it's the only way to use rsync.

Backup jobs can't be modified to point to a USB port while the original external share drive is not attached. They can however be deleted while the drive is not attached. So if I want to keep the jobs, which I do, I have to put up with the failures until I can retreive the original hard drive.
Presently I schedule several jobs to run (weekly) in the early morning and have the last job auto eject so that I can just disconnect and store the drive. It would be nice to just be able to change the schedule when I am next on the computer.
