Are the drives the same make/model and running the same firmware?
You could try increasing the Spindown time to something like 30 minutes and see if that has an effect. Otherwise, power down the NAS normally and remove the drive and Run SeaTools on it. That will give you a thorough diagnosis of the state of the drive. Be prepared for a bit of a wait though, as the Disk surface scan in SeaTools is mind numbingly slow in my experience.
Remember to mark which bay the drive came from too and don't get that order mixed up.
