Bad flash would be pattern 4. That says that the ReadyNAS flash memory has issues, not the the hard drives. It seems unlikely, but I wanted to confirm the LED pattern (since you didn't sound certain).
The idea about booting with one drive at a time is simply because sometimes a failed drive will prevent the NAS from booting. This was an alternative to linux reader, so if you get your data off with that you don't really need to do it. However, some of them should boot by themselves (the ST31000524AS should result in a factory install).
The vendor tools warn you if you try to run a destructive test (and you shouldn't tun those destructive tests before you get the data back).
