Question about ISCSI and Plex?
So, I just bought a ReadyNAS 312 this weekend. I set it up and everything seems to be working great so far.
What I am wondering is if there is any way to access the data that is added through ISCSI in the Plex Media Server app?

You could access that data using Plex Media Server running on the PC on which the initiator mounts the target, but not on the NAS itself. Only mount the target on one PC at a time. You could share the files on the target from the PC you mount it on.

I've experimented with PLEX is many different configurations, I find iSCSI a little slow for this purpose, I get almost double the throughput when using SMB.
