USB backup knocks ReadyNAS offline

Is 6.4.1 the most recent firmware? If so, that is what we have. I checked for updates this morning to make sure there wasn't something different.

6.4.1 is the most recent - it was released on 11/30/15. 6.4.0 came out in October.
You might want to check the SMART stats on the internal hard drives, and perhaps run a diag on the USB drive too.
I also suggest creating a new backup job, and see if that works correctly.

Thanks, I will try creating a new backup job. The drives all appear to be in good working order and I am currently trying a brand new external drive today but that didn't solve the issue either.

If that does not resolve it you may wish to try downgrading to 6.4.0 to see if that resolves the problem.

download the zip from http://kb.netgear.com/app/answers/detail/a_id/29938/~/readynas-os-version-6.4.0 and extract the image file.
Then install it using the web ui (system->settings has a manual install control).
