No. It's just another protocol as far as the NAS is concerned.
Details on setting up rsync depends on the firmware you are running - can you let us know what both duos are using?
Also, I am thinking your goal is to set up rsync backup (perhaps on a schedule). Is that correct?

The unit called "NAS" is a ReadyNAS Duo with S/N 1VB384RK015FD, firmware RAIDiator 4.1.14 [1.00a043]. At now, it is used at 9% of 1 TB
The unit called "NAS_BKUP" is a ReadyNAS Duo with S/N 24M30B7G001BF, firmware RAIDiator 4.1.14 [1.00a043]. At now, it is fully available.
My goal is to have a full-time working and synchronized copy of the whole content of the "NAS". I use this unit as "file server" of my office, available to all the users (me and my two collaborators). At now, I didn't feel the need of a backup copy (I have two mirroring disks in the unit!), but...
Recently, a strange issue occurred - two times - to the "NAS" unit (no more on-line in the LAN, powered-on but not responding at all, even by pressing the power on/off button to turn it off).
To avoide a disaster, I should like to have a second mirroring unit, which is on-line all the time with the same content of the first unit. This should avoid downtime to my office, needed to move the two disks from one unit to the other (assumed that this scenario should be safe for my data).
Is this purpose of mine in the possibility of the Rsync protocol? Could I obtain a "real-time" backup service between the two units, while I'm normally using my file server (opening/moving/deleting files)?

You should have a backup of the NAS, as RAID is not enough to keep the data safe.
Rsync backup can copy your shares from NAS to NAS_BKUP on a schedule. It is not continuous, but it can be as often as once per hour.
