I may be totally out of date here but, as far as I understand, the 6TB WD Red HD is not on Netgear's hardware compatibility list for your ReadyNAS NV+ v2 but the 4TB WD RED (WD40EFRX) HD is: http://kb.netgear.com/app/answers/detail/a_id/20641/~/hardware-compatibility-list-hard-disks
And so, I don't believe that Netgear or anybody else can guarantee that your 6TB HD will work with your legacy NV+ v2.

Netgear will almost always deny support if the disks you are using are not on the HCL for your NAS.
Having said there, there is no obvious technical reason why this disk shouldn't work. But Matt is blazing a new trail - at least no one else has posted anything about using this drive in a v2 NAS.
