If you want to remove the existing two drives while you are testing, power the NAS down and remove them. Though perhaps not strictly necessary, it is good to label them so you can return then to the same slots.
Then install the one you want to test and power up.
Be careful to return to power down the NAS before you reinsert the old drives. Make sure they are the only drives installed. Then power back up.
Any hot-insertion is treated by the NAS as a new drive (e.g. it runs a brief diag and then wipes it).
