You could try renaming the users then renaming them back.
If that does not resolve it perhaps try 6.4.2 Beta?: ReadyNASOS 6.4.2-T59 (Beta 1)Did you change the NAS hostname a while back?

Nope... updated smbpasswd, did reload on smb.service (since there are active connections, didn't want to restart it), and tried smbclient on problem user. Still get access denied.

Nope, haven't changed the hostname since I first installed the 516. Renaming the user did resolve the issue. I do wonder why though. :) As long as it's working, I'm a happy clam. Thanks!

We have reproduced a problem with accessing home shares and have a fix in ReadyNASOS 6.4.2-T72 (Beta 2)
Most likely you changed the hostname of the NAS after the user accounts were created.- microdev2Jan 11, 2016Aspirant
I also have not renamed the host and same problem happened with me. Renaming the user back and forth fixed it.
