You can try running vendor diags (seatools for seagate, lifeguard for western digital) - which I think will confirm that the drive is dead.
If so, you probably will need to go to your old backup.
One thing to try: if you put in just disk 2 (in slot 2 with the NAS powered down), does the NAS boot?
