I'm not sure which line in the config is causing the 'unknown option' message offhand, but you'll need to stop the ReadyDLNA service before running it manually. You can either use Frontview or the following command:
kill `cat /var/run/minidlna.pid`
kill `cat /var/run/minidlna.pid`
