Well.... this could depend on a few factors. But I can confirm that the Samsung PLEX app does tend to lean on DirectPlay when you're on the same LAN. This was a bother for me due to certain aspects of our home networking menagerie, so I had to force the app *not* to DirectPlay in my case.
The upside of this, though, is that if the Samsung device is likely to be the only one you use for playback *and* you have decent bandwidth, then much of your media should play fine. BUT, if the app drops out of DirectPlay for any reason you'll be dead in the water, due to the lack of transcode in 1xx boxes. I've heard 3xx boxes can do OK, but you'd really want a 5xx or 7xx for best results. Same will be true if you start viewing your PLEX-based media from other devices like tablets, phones, web...
