You need to enable rsync for the USB disk.
Use the IP address of the NAS, not the name.
You don't need the login details.
Remove deleted files on source will remove files on the backup ( destination) that are no longer on the source ( your NAS). It will not delete anything from the NAS.
Exclude folders will exclude all folders you list, separated by commas. There is a particular syntax for entering folder names with spaces. My advice would be to setup separate rsync backup jobs for each share that you have and exclude snapshot folders. Much more versatile than rsync of a whole volume.
I don't think you can automatically unmount the USB drive at present, you'll need to do it manually using the GUI.
