Update: So my orginal drive in slot one was dead - post removal testing and it won't even spin up. Log indicates multiple errors in the final days and apparently my email notifications were disrupted at some point and the reason why I didn't have any warning.
So procedure was I shutdown, removed the dead drive then restarted. Performed a USB backup via direct connect to the ReadyNAS and using the RAIDar browser interface - 1 TB took almost 48 hours so next time maybe I do that straight over ethernet. Added a new drive to the open bay and now system s fully redundant again.
Thank-you community for helping me walk through this. I wish there was more information in the Netgear procedures about how redundancy works when the NAS is live vs shutting down and rebooting. Most of the info I could find was from threads in community and there were contradictions between various NAS products discussed here.
Thanks again everyone!

I'm glad it worked out.
I do want to add that many v1 owners mistakenly think they have a v2. If your duo is running 4.1.x firmware, then it is a v1. More information on model identification (and the labeling confusion) is here: http://www.rnasguide.com/2012/01/09/how-to-tell-whether-i-have-a-duo-v1-or-duo-v2-or-nv-v1-or-nv-v2/
