Share the ReadyNAS folder on the PC, and drop the U$ in the backup job configuration.

I would like to use the built in Administrator share u$ if possible.

If I go the opposite way from MyPC/u$ to the ReadyNAS it works fine.

Why?
FWIW, you could alternatively access the share using u$ from another Windows PC in file explorer, and note the full path name there.

U$ is a local variable on the PC. The NAS has no idea what it's value is and it does not pass in a Linux based backup job as a variable name the PC can fill in. So, it's taken literally. You need to replace it with it's value on the PC. StephenB has suggested one way to figure that out.
