Create cloud backup of server backup stored on NAS

I want to send my server backup which is stored on my ReadyNAS in a LUN to Amazon S3 as an off-site backup alternative to my local NAS backup. From my NAS Cloud tab, I am only able to backup folders in my NAS "data" directory. I am unable to browse to my mapped drive containing my server backup.
I have Windows Server 2016
Using built-in Windows Backup to backup my server's C drive and my System State
Backup size: 1 TB of data to a LUN
LUN mapped to drive E on my server.
How can I access that LUN on my NAS to send it to the cloud?

You can't do it from the NAS side. An iSCSI LUN is, to the NAS, mostly just a reserved block of space. It is unaware of the file system structure within it. You will have to do the backup from the initiator side.
If you want the backup capability, you need to save the data in a share, not a LUN.
