Unfortunately, it's impossible for anyone to provide you with a definitive recommendation. The construction of your house, the presence of nearby radio interference and other Wi-Fi networks will determine whether you can make do with just the C6300 or whether you'll need extenders on the top and bottom floor.
Certainly, central placement of the C6300 on the middle floor will give it the best chance of covering the whole residence. If your house is wired for Ethernet, then you can take advantage of it by deploying wired Access Point strategically in areas with poor coverage. You can also use Powerline as a substitute for Ethernet, although results will be less reliable; it may work great or not at all.
Start with just the C6300 and see how it goes. Map out the dead spots where you want to boost the signal. It should become evident whether you need 1 or 2 range extenders.
