Home Folders disappeared after updating to 6.10.6
I have upgraded my ReadyNas to 6.10.6 to fix the issue with Antivirus, but after update the home folders went offline, and when I check home folders option, it shows "not shared". SMB Share is enable (On) but still shows are "Not Shared".
Please help.

First I'd see if you can access /home (and its subfolders). You can do that from a Windows PC by accessing the data volume (called data if you run xraid) with the NAS admin credentials.
For instance, run CMD and enter
net use * /delete /y net use t: \\nas-ip-address\data /user:admin nas-admin-password
using the real nas ip address and admin password of course. Be careful with the typing, especially the two different slash directions.
This should mount the full data volume as drive letter T. You can then access t:\home and see the home folders and their contents.
If the contents are there and you don't have a backup, then I suggest backing them up before proceeding further.

I ran it, and yes, I can see the \home folder and all of its contents. I was also able to browse HomeFolder's contents using Nas admin page.
I should say that in the same NAS we have 3 other shared folders and they are working properly in the network.
Q- Should I back up only home folder contents or the entire NAS?
and what are the next steps to fix it.

By the way when I try to access \\nas-IP-address, it shows me all the individual share folders for users (user home folders), but gives the error (you do not have permission to access). Looks like the issue is only with the home folders for each user due to their splicit permissions settings.
Does this give any idea?
