Thanks siigna & mdgm
I'm not sure of the implications of mdgm's post but here's what I've done - I backed up Users and groups, then opened the zip file in 7-Zip. I double clicked the /etc/shadow file to open it in Notepad (I can't get it to open in Notepad++ unless I extract the file and after mdgm's post I was trying to avoid doing that) and replaced the root: line as far as the 3 colons - Notepad doesn't separate the lines well.
I then saved and closed shadow, closed 7-Zip and restored the backup. The readyNAS wanted to reboot so I let it. When it rebooted I did another backup and checked that the line had indeed replaced. I also extracted shadow file to my desktop and checked it in Notepad++. Everything looks OK, the root line is excatly the same as the one siigna gave me.
Unfortunately still getting Access Denied when I try to log in as root
