a) How long will it take my ReadyNAS to sync up each new red drive?
Smallnetworkbuilder.com measured 4 1/4 hours per TB. Note disk fullness doesn't matter for sync time.
Assuming that's still the case with the current firmware, the first one would take about 8-9 hours, and the second one 16-17. I believe there will need to be a reboot in the middle of the second sync (to trigger the vertical expansion).

b) Is hot swapping my drives recommended or should I switch my ReadyNAS off before swapping my 2TB drives out?
I always recommend hot-swapping - that ensures that the firmware detects the drive insertion.

Hi StephenB
I understand and accept your advice, I just have one query when you said the following: "I believe there will need to be a reboot in the middle of the second sync (to trigger the vertical expansion)". Well, I don't recall ever reading this in the Duo V2 manuals/user guides (even after a quick check). All the manuals say is that I will get an email notification when syncing is complete, but that in itself is not important, can you say whether I will get an email notification when the second sync reboot is required?

I don't think so (though I am not a v2 owner).
Perhaps insert the second disk in the morning, and check the system early evening.

You shouldn't reboot during a resync. If expansion doesn't take place after the resync to add the second higher capacity disk completes then you could try a reboot, wait 5-10 minutes and if expansion hasn't started perhaps try one more reboot.
