Going forward you would only need to install firmware updates once as usual.
In all aspects with the possible exception of the uboot update (which may/may not work from just the one update) installing 5.3.5 once would give you all the benefits of it. The OS on the flash is updated completely as is the OS on the disks. In my view it's highly unlikely that further uboot updates will be needed in the future for the Duo/NV+ v2 and if there are updates there shouldn't be major problems that stop the NAS from booting with not being on the latest.
