How to make an exact copy of a share using backup or rsync

It compresses the data flowing on the network connection. It is not compressing the files. If the backup is on the local lan, I'd just leave it off.
With rsync in particular, there's a backup option that does what you want. It's called "Remove deleted files on source.", and is a check box in the "step 1" pane.
For other protocols, that option isn't available. You can choose to check the "Remove the contents of the backup destination before a full backup is performed." option in step 4 (at the bottom), and schedule an full backup from time to time. The risk is that main NAS happens to fail in the middle of the full backup, you'll lose data.
