Yes.
Note some of the rackmount chassis can't handle > 2 TB disks in every slot (a hardware limitation). But that's not an issue with the RN4200v2. So you should upgrade the firmware - that will let you use the larger disks.

And since you've already done a factory default, now would be the time to consider moving to OS6 on your 4200V2. It's not officially supported by Netgear, but it works and aleviates several problems related to the old Debian distro OS 4.2.x is based on.

The RNDX4420 is a desktop 4-bay ReadyNAS, the NVX that shipped with 4x2TB disks.
The NVX can’t run OS6 as it has a 32-bit CPU.
