M4100-26G DHCP Relay with VLANs

Regarding enabling it on the ports as well: No, if you got it enabled on the VLAN there is no need to enable it on individual ports.
Regarding routing: For every VLAN you want the switch to do DHCP relaying you also need an IP address assigned on the switch. Without an IP address the switch is not able to communicate with the DHCP clients on the individual VLANs.
