Hello gjj2017,
Let's start with what ReadyNAS model you are using and what firmware version. Did you also try other USB port on the NAS and please post here the complete backup job logs.
Welcome to the community!

Model: ReadyNAS NV+ [X-RAID]
Serial: 21S195RX0051C
Firmware: RAIDiator 4.1.5 [1.00a043]
Memory: 256 MB [2.5-3-3-7]
IP address: 192.168.0.100
Volume C: Online, X-RAID, 2 disks, 35% of 456 GB used
Have not tried the other port.
There really is no further info in the backup log. It is a law office and the filenames contain case names that are private that is why I sent only a subset.
Here is the header and top few lines:
Error encountered copying data from source path ==> /USB_HDD_4/RoehlBU due to unknown reason. Please see log.
cp: cannot create regular file '/USB_HDD_4/RoehlBU/./Taxes/2017 Taxes/K-1 Forms 2016/K-1 Roehl\'s Cars Roehl\'s Prop - Taylor.pdf': Operation not supported
cp: cannot create regular file '/USB_HDD_4/RoehlBU/./Taxes/PIT-8453.pdf': Operation not supported
cp: cannot create regular file '/USB_HDD_4/RoehlBU/./Antelope Point/receipt_2017-04-13_092633.pdf': Operation not supported
cp: cannot create regular file '/USB_HDD_4/RoehlBU/./Antelope Point/receipt_2017-04-13_091714.pdf': Operation not supported
cp: cannot create regular file '/USB_HDD_4/RoehlBU/./Antelope Point/receipt_2017-04-13_092049.pdf': Operation not supported
Then there are many more file names listed after that with the exact same error then the log ends...............no other pertinent info, in fact that is all she wrote.
Thanks,
Glen

I suspect you've already checked this, but just in case... Have you confirmed that you can copy these files from the NAS to a PC (drag and drop)?
Also, are some files being backed up? Or are all the copies failing?

Yes, I have copied those files to a PC. I have also been able to connect the drives in question and copy files to and fro.
As to "are some files bieng backed up?" - I do not believe that any files are getting copied. I created a couple of test backup jobs with 6 or 7 files in a folder and all of those fail as well.
I have the one drive that we use on Tues and Thursday (which, after chkdsk /f, worked last Thursday) attached to the NAS tonight. Will see if it works a second time.
One thought I had: When the drives are swapped out each night, we just detach them from the NAS, we do not go into FrontView and disconnect them first. While this has not been a problem for a couple of years, might doing that over a very long period of time cause this issue?
Thanks.

Tueday Night Update....
The NAS ---> USB backup to the Tuesday drive worked this evening. Will try the other one Wednedsay night and see.
To reacp: This above mentioned drive that was successful tonight was not for the past few weeks. After disconnecting it via FrontView and performing a chkdsk /f on a PC, it has been successful 2x's
The other drive, same steps, disconnect, chkdsk /f. A test backup worked on it but the scheduled one subsequently failed. 2x's
Both drives performed as expected when connected to a PC: read, write, erase, chkdsk ok.
Will report back Wednesday evening.
Thanks,
Glen
