Nighthawk X6S AC3000 Router
I have a USB hooked up as a ready share drive
It mounts over the network and the files are visible but when I try and open them it says the file is not available

Problem Resolved. I switched to a 4TB external drive and it worked fine

What is this "USB"? A disk drive of some sort? USB stick?

Is this over your LAN? Is it plugged into the router's USB port using ReadyShare?

The model is the R7900P. The wifi speed is 802.11AC but the computers are connected to the router through a gigabit ethernet backbone.
The USB drive is a Seagate Backup Plus 5TB drive plugged into the routers USB Port using readyshare

Greetings. These are the questions we would typically request answers for in this situation:
-How is the disk formatted?
-Does the disk have multiple partitions?
-Does this behavior happen when another drive plugged in?
-Has the router been updated recently?
-If so, how was the update performed? (Auto Check and Update, Genie, Manually)? Manual update continues to be the most reliable method.
-Did you apply any saved .cfg files from an earlier FW?
-Has the router been reset to defaults and tested with the disk?
Optional:
Consider trying another version of FW.

That's bigger than anything I have seen reported as RapidShare compatible.

Past experience here suggests that firmware, formatting and whatever else you try will do nothing to enable this feature in many/most big drives.
